摘要
目的研究土鳖虫含药血清对肝癌HepG-2细胞增殖的抑制作用及其对细胞凋亡和细胞周期的影响,探讨土鳖虫抗肿瘤效应。方法以土鳖虫乳剂给SD大鼠灌胃,采血并制备血清。四氮唑盐法(MTT法)检测含药血清对肝癌HepG-2细胞增殖的抑制作用,流式细胞仪测定细胞周期和细胞凋亡。结果土鳖虫含药血清可明显抑制肝癌HepG-2细胞体外增殖,20%土鳖虫含药血清作用72h后抑制率高达56.728%,抑制率呈浓度依赖关系。流式细胞仪检测细胞凋亡发现肝癌HepG-2细胞以坏死居多,细胞周期多阻滞于G0-G1期。结论土鳖虫含药血清对肝癌HepG-2细胞体外增殖有明显的抑制作用。
Objective To investigate the inhibitory effect of serum containing eupolyphaga on the proliferation of human hepatoma cell line HepG - 2 and its influence on cell apoptosis and cell cycle in vitro. Methods The rats were administrated by gavage and then the blood was collected to prepare serum containing eupolyphaga. The MTT. assay was used to examine the proliferation inhibitory activity and the flow cytometry was used to assay the apoptosis and cell cycle. Results The serum played obviously anti - proliferation activity on HepG - 2 in vitro. The inhibition ratio depended on the serum concentration. The inhibition rate was up to 56. 728 % after treatment with 20 % serum containing eupolyphaga for 72 hours . The results of the flow cytometry revealed that most of HepG - 2 cell was necrotic and the cell cycle was blocked in Go - G1phase after pretreated with the serum. Conclusion The serum containing eupolyphaga has an obvious inhibitory effect on the proliferation of HepG - 2 in vitro, and eupolyphaga emulsion as a new antineoplastic herb preparation is worth of further study.
