芳维A酸氨丁三醇对Hacat细胞信号转导途径的影响

摘要 目的通过研究芳维A酸氨丁三醇(Arotinoid Trometamol,AT)诱导HaCat细胞后的基因表达差异,初步探讨AT治疗银屑病的作用机制。方法应用基因芯片技术,检测AT(10-6mol/L)诱导HaCat细胞24小时前后基因表达谱的改变。结果在4000条人类全长cDNA中,有580条的表达量差异达到2倍以上,其中,注释完整的基因有253条(143条表达量增加,110条表达量减少)。差异表达的基因主要是细胞内信号基因(17.8%),细胞受体基因(8.3%),DNA结合、转录因子基因(11.9%),细胞因子基因(4.0%),细胞周期及细胞凋亡的调控基因(5.6%)。结论AT能够通过影响信号转导途径中包括KLF4、HSPA8、HSPCB在内的多个基因表达而发挥其生物学作用。 Objective To investigate gene expression pattern of HaCat cells after AT treatment. Methods Applying cDNA microarray to identify differentially expressed genes in HaCat cells after AT treatment for 24 hours (10^-6mol. L^-1). Results Among the 4000 full -length human cDNA, 580 genes exhibited differential expression ( ≥2 folds) ;253 genes with complete annotation were also identified, including 143 genes upregulated and 110 genes down - regulated. The differentially expressed genes belongs to catalogs of introcellular signaling transduction pathway, receptor genes, DNA binding, transcription factors , cytokine genes, cell cycle and apoptosis genes. Conclusion Microarry technology can help researchers to understand the AT induced gene expression pattern in HaCat cells in a high through - put way. We find many cytokines of signal transduction, like KLF4, HSPA8 and HSPCB, can be regulated by this kind of drug which is used in treatment of psoriasis.
