摘要
目的:观察自发性高血压大鼠(SHR)肾脏α-addu-cin基因表达增龄性改变的一般规律及高盐摄入对其的影响.方法:以雄性SHR为观察对象,按周龄分为幼年组(9wk,13只)成年组(28wk,15只)SHR和老年组(53wk,13只).应用实时荧光定量PCR进行α-adducin基因表达的检测.结果:成年组与幼年组SHR相比,肾脏α-adducin基因表达无统计学差异(P>0.05);而老年组肾脏α-adducin基因表达比成年组(0.37±0.12vs5.43±2.22,P<0.05)和幼年组(0.37±0.12vs4.48±1.30,P<0.05)降低,均具有统计学意义.在各年龄组SHR中高盐组和正常盐组比较,肾脏α-adducin基因表达无统计学差异(P>0.05).结论:SHR肾脏存在增龄性α-adducin基因表达异常;短期高盐摄入对α-adducin基因表达无明显影响.
AIM: To examine the alteration of α-adducin gene expression in the kidney of spontaneously hypertensive rats(SHR) with aging and to evaluate the effect of high sodium loading on it. METHODS: The expression of α-adducin mRNA in the kidney of different age groups of male SHR (9, 28 and 53 wk ) was detected using relative quantitative real-time polymerase chain reaction. RESULTS: α-adducin mRNA expression in the kidney was not significantly different between 9-week-old and 28-week-old SHR( P 〉 0. 05 ). Nevertheless, it was significantly lower in the 53-week-old male SHR than in 9-week-old SHR (0.37 ±0.12 vs 4.48 ± 1.30, P 〈 0.05 ) and 28 -week-old SHR (0.37 ± 0. 12 vs 5.43 ± 2.22, P 〈 0. 05 ). No significant difference was found in the α-adducin mRNA expression between the low-salt and highsalt swbgroups in each group ( P 〉 0.05 ). CONCLUSION:α- adducin mRNA level in the kidney of SHR decreases abnormally with aging. Short-term high-salt loading exerts no influence on α- adducin mRNA level in the kidney of SHR.
