摘要
为建立高山红景天试管苗的缓慢生长法保存体系,在低温、弱光及不同浓度甘露醇条件下对两种来源的试管苗进行缓慢生长法保存,并对其恢复生长植株的DNA含量进行分析.结果表明:低温(6±1)℃和弱光600 lx(8 h·d-1)条件下,试管苗在添加有甘露醇10 g·L-1的1/2MS+蔗糖20 g·L-1+琼脂7 g·L-1培养基中可保存18个月,平均存活率在85%以上,且恢复生长后试管苗长势良好.上述方法保存后恢复生长的植株经流式细胞仪检测表明核DNA含量没有发生改变.该结果为高山红景天种质资源缓慢生长法保存的有效性和可行性提供了理论依据.
In order to establish conservation system of in vitro shoots of Rhodiola sachalinensis by slow growth, studies on conservation of in vitro shoots of two resources by slow growth under low temperature, week illumination and mannitol of different concentration were carried out, and DNA contents of those recovered plantlets were analyzed. The results indicated that more than 85 % of in vitro shoots conserved on 1/2MS supplemented with 20 g· L^-1 sucrose, 7 g· L^-1 agar and 10 g·L^-1 mannitol maintained viability after 18 months under (6±1)℃ and 600 1x conditions, Both nuclear DNA contents maintained as normal, Thus, it was suggested these procedures would be effective and viable to conserve Rhodiola sachalinensis by slow growth as above.
