摘要 目的:通过对临床子宫内膜异位症(endomotriosis,EMS)患者异位内膜及在位内膜Survivin mRNA及Cox-2mRNA表达测定,探讨Survivin及Cox-2在EMS发病及发展中的分子生物学机制及其两者的相关性。方法:随机选取开腹手术或腹腔镜手术EMS患者异位内膜组织、在位内膜组织和正常妇女(除外激素依赖性疾病或激素治疗者)正常子宫内膜各20例,应用半定量逆转录-聚合酶链反应(RT-PCR)方法,测定Survivin及Cox-2的mRNA表达水平。结果:与正常妇女子宫内膜相比,EMS患者异位内膜的Survivin mRNA表达水平上调,Cox-2mRNA表达水平也明显上调,且总体Survivin及Cox-2mRNA表达与EMS呈正相关,两者有明显的相关性。而EMS在位内膜与之相比,Survivin上调,Cox-2变化无统计学差异。结论:EMS患者Survivin及Cox-2上调可能是EMS发生、发展的分子生物学机制之一,与异位内膜的发生和种植有关,且两基因在疾病的发展中关系密切。 Objective: To evaluate the molecular mechanisms by measuring the gene expression of Survivin and Cox-2 which occur in patients with endometriosis (EMS) and to investigate their effects on the initiation and maintenance of EMS. Methods. The ectopic endometrium and eutopic endometrium were obtained from 20 patients with EMS (cases) and from 20 normal endometrium. The mRNA expression of Survivin and Cox-2 were measured by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results: Compared to normal endometrium, the mRNA level of Survivin of ectopic endometrium and eutopic endometrium significantly increased, the mRNA level of Cox-2 was significantly increased also; The total mRNA level of Survivin and Cox-2 was negatively correlated with EMS, and their correlation was obvious. Conclusion: EMS is predominantly accompanied by abnormal regulation in the mRNA expression influencing the Survivin and Cox-2, which are associated with occurrence and development of EMS and may influence the initiation and maintenance of EMS.
