摘要
目的:探讨叶绿素显示液对狗胃淋巴结显色的应用价值。方法:13只成年狗分为A:对照组(1只)和B:实验组(12只),B组于胃壁内注射叶绿素显示液,观察胃16组淋巴结的染色情况,并在不同时间(1、2、4、8、24h)取显色淋巴结作光、电镜观察;从狗股静脉采血,测定血清中叶绿素的含量。结果:注射叶绿素显示液后,狗胃各组淋巴结很快显色(1~5min)。光镜下观察到叶绿素进入淋巴结的被膜下窦和小梁周窦。电镜下观察,淋巴结皮质、髓质细胞内、单核细胞和巨噬细胞内可见叶绿素颗粒;叶绿素在血液中的浓度,4h最高,为350.46μg/ml;之后随时间变化逐渐减少。结论:(1)叶绿素显示液是一种理想的淋巴染色物质,染色迅速,与周围组织对比明显,且染色后无不良反应和并发症。(2)经胃壁内注射叶绿素显示液,能够有效显示胃的各组淋巴结,可提高胃淋巴结的清除率及避免损伤周围组织。
Objective: To investigate the value of chlorophyll for disclosing lymphonodi gastrici of dogs. Methods: 13 dogs were divided into two groups: control group (A) with one dog and experimental group (B) with 12 dogs. Staining results of 16 groups of lymphonodi gastrici were observed by naked eyes after injecting the disclosing solution into gastric wall, and then, tissue structure and ultrastructure of lymph nodes, taken out at different time (1, 2, 4, 8, 24 h ) after staining, were observed under light and electron microscope. The concentration of chlorophyll in the blood was measured by biochemistry technique after collecting venous blood from femoral vein of dogs. Results: All lymphonodi gastrici were stained quickly ( 1 ~5 min) after injection. Chlorophyll grains were found in subcapsular sinuses and trabeculum sinuses, and were visible in cortex nodi lymphatici cells, lymphomonocyte and macrophagocyte. The concentration of chlorophyll in the blood reached the peak of 350.46 μg/ml at the 4th hour after injection, and then decreased gradually, Conclusions: ( 1 )The chlorophyll disclosing solution was an ideal lymph dye, for its rapid staining ability, well contrast with surrounding tissues and none of toxic symptoms or complication. (2)All groups of lymphonodi gastrici can be showed efficiently. The method may improve the clearance rate of stomach lymph nodes and avoid injuring adjacent tissues.
