脑膜瘤中凋亡相关基因Survivin和p53的表达研究

摘要 目的探讨凋亡相关基因Survivin和p53在脑膜瘤中的表达及其相互关系。方法应用免疫组化SP法,检测86例脑膜瘤组织、15例正常脑膜组织中凋亡相关基因Survivin、p53蛋白的表达,并统计学分析两者之间的关系。结果Survivin在正常脑膜组织和脑膜瘤组织中阳性表达率为0%和61.6%,两者比较具有显著性差异(P﹤0.001)。Survivin表达阳性率在脑膜瘤II级(76.5%)和I级(48.2%)之间、III级(93.3%)和I级之间差异具有统计学意义(P﹤0.05和P﹤0.01)。p53在正常脑膜组织和脑膜瘤组织表达的阳性率为0%和36.1%,两者比较具有显著性差异(P﹤0.05)。p53表达阳性率在脑膜瘤II级(52.9%)和I级(20.4%)之间、III级(73.3%)和I级之间差异具有统计学意义(P﹤0.05和P﹤0.001)。Survivin与p53蛋白表达共阳性27例,共阴性29例,呈显著正相关(rs=0.687,P﹤0.01)。结论(1)Survivin和p53在脑膜瘤中的表达与其恶性程度明显相关;(2)Survivin基因过度表达和抑癌基因p53失活在脑膜瘤的发生和恶化中起协同作用;(3)Survivin将是脑膜瘤免疫治疗理想和有效的靶基因。 Objective To explore the expression of apoptosis related gene Survivin and p53 in meningiomas and the correlation between them. Methods Using the SP (streptavidinbiotin peroxidase) method, 86 specimens and archived paraffin-blocks of meningioma and 15 normal meninges were detected for the expression of apoptosis related gene Survivin and p53, and the interaction between them were statistically analyzed. Results The positive rates of Survivin expression in normal meninges and meningiomas were 0 and 61.6% (P 〈 0.001). Survivin positive immunoexpression rates were 48.2%, 76.5% and 93.3% in meningiomas of grade Ⅰ, grade Ⅱ and grade Ⅲ. The positive ratios of Survivin showed a significant difference between in grade Ⅱ and in grade Ⅰ (P 〈 0.05), in grade Ⅲ and in grade Ⅰ (P 〈 0.01). The positive rates of p53 expression in normal menings and meningiomas were 0% and 36.1% (P 〈 0.05). p53 positive immunoexpression rates were 20.4%, 52.9% and 73.3% in meningiomas of grade Ⅰ, grade Ⅱ and grade Ⅲ. The positive ratios of p53 showed a significant difference between in grade Ⅱ and in grade Ⅰ (P 〈 0.05), in grade Ⅲ and in grade Ⅰ (P〈 0.01). There were 27 patients who had both Survivin and p53 expression at the same time, 29 patients had negative expression of them. A positive correlation (rs=0.687, P 〈 0.01) between Survivin and mutation p53 expression was found. Conclusion ( 1 )The expression of Survivin and p53 in meingiomas was obviously associated with its malignancy. (2)The over expression of Survivin gene and de-activation of antioncogene p53 play synergetic roles in the process of oncogenesis and malignancy of meingiomas. (3)Survivin may be an ideal and effective immunotherapeutic target gene in meingiomas.
