α-硫辛酸对2型糖尿病大鼠血清和肝组织氧化应激及脂质代谢障碍的改善作用 被引量:19

摘要 目的观察α-硫辛酸对2型糖尿病大鼠体内氧化应激状况及脂质代谢障碍的改善作用。方法10只雄性大鼠随机分为2组:糖尿病对照组和糖尿病硫辛酸治疗组,健康Wistar大鼠4只为正常对照组,共观察12周。测定各组大鼠的血糖、血清及肝匀浆中的氧化还原系统指标及脂质谱水平。结果糖尿病对照组血清和肝匀浆中的抗坏血酸、谷胱甘肽、维生素E、过氧化物歧化酶和丙二醛水平较正常对照组明显异常(P<0.05和P<0.01),经α-硫辛酸治疗后有所改善(P<0.05和P<0.01)。糖尿病对照组血清和肝匀浆中的总胆固醇、甘油三酯和游离脂肪酸水平较正常对照组增高(P<0.05和P<0.01),α-硫辛酸治疗后有明显降低(P<0.05和P<0.01)。结论α-硫辛酸能改善2型糖尿病大鼠体内的氧化应激状态,改善胰岛素抵抗,进而改善脂质代谢紊乱。 Objective To investigate the effect of α-lipoic acid on oxidative stress and changes in lipid profile in type-2 diabetic rats. Methods Four healthy Wistar rats were taken as normal control group and 10 diabetic rats were divided randomly into two groups: diabetic control group and lipoic acid-treated diabetic group. Malonaldehyde (MDA) ,antiscorbic acid (VC) ,vitamin E (VE) ,glutathione (GSH) ,superoxide dismutase (SOD) ,total cholesterol (TC) ,triglyceride (TG) and free fatty acid (FFA) in serum and liver homogenate were measured. Results There were significant differences in levels of VC,GSH,VE,SOD,MDA and lipid profile in serum and liver homogenate between diabetic control group and normal control group (P 〈 0.05 and P 〈 0.01 ). The measurements of parameters were improved in lipoie acid-treated group ( P 〈 0.05 or P 〈 0.01 ). Conclusion Antioxidant α-lipoie acid can improve the oxidative stress and the dyslipidioses in diabetic rats possible by preventing insulin resistance.
