摘要
采用高脂饲喂中国实验用小型猪模型,观察乙酰胆碱(Ach)拮抗剂山茛菪碱(654-2)对培养冠状动脉内皮细胞(EC)代谢、增生、血小板生长因子(PDGF)基因表达的影响,探讨ACh在冠状动脉粥样硬化发病机制中的作用。结果发现:1.654-2可明显减缓EC的生长、传代;2.654-2使EC的PDGF-A、B链基因表达降低;3.654-2增加EC的NO和还原型谷胱甘肽GSH水平、减少过氧化脂质(LPO)产生。
This study investigated the influence of anisodamine-an acetylcholine antagonist on metabolism, proliferation and PDGF gene expression of coronary artery endothelitun cell (EC) ofminipig subjected to high cholesterol diet and found that (1) EC growthing more fast in both controland amlodipine groups with most of them in S+G2+M phases compared with that of anisodaminegroup (P<0.05); (2) PDGF-A、B mRNA showing lower level in anisodamine group; (3) lipidperoxide(LPo) and lactic acid (LA) producing lower in anisodamine group (P<0.05) with a higherconcentration of nitric oxide (NO) and increased glutathione (GSH).
