摘要
用2%多聚甲醛、2%戊二醛配伍不同浓度的单宁酸(0.5~5%)媒染固定液灌流大鼠,取脑切片,放入2%氯化铁溶液中呈色,成功地显示了脑内各级血管及毛细血管网的形貌.高倍镜下可见各级血管呈空心管道状,其壁为多条细纤维丝状结构编织或呈同心圆状排列而成,具有较强的立体感.血管间的交又、重叠、吻合均清晰可见.结果证明:含2~3%单宁酸的媒染固定液媒染效果最好.
The rats were perfused with compound staining-fixative solution of 2% paraformaldehyde, 2% glutaraldehyde and various concentration of tannic acid(0.5-5% TA). The specimens from the brain were cut into slices and immersed in 2% ferric chlorid solution to reveal colors by che-mical reactions. The capillary network and arterioles were well manifested as vacuous tubes under light microscope- The wall of the vessels was formed by many woven or concentrically arranged slender fibers, creating distinct three-dimensional sensation. The crossing, overlapping and anastomosis of capillaries and arterioles were be seen clearly. The findings suggest that the effect of staining was best with the 2-3% concentration of tannic acid. The study provided an ideal, simple and direct method for further investigating microvasuclar and pathololgical changes.
