摘要
目的:探索建立一个新的闭胸式犬慢性心肌缺血和梗塞模型.方法:选用15条健康杂种犬.经右股动脉送入自制75%或50%缩窄器,造成左旋支(LCX)或前降支(LAD)狭窄.术后5~10月,9条犬进行选择性左冠状动脉造影,左心室造影及心肌病理学检查.结果:LCX闭塞犬5条;95%狭窄3条;LAD95%狭窄1条.整体射血分数下降(P<0.05).在LCX病变组,左心室膈段及后基底段运动消失5条;运动减弱3条.心肌间质结缔组织增牛,钙化灶形成,T管扩张及区域性心肌溶解.结论:为进一步研究心肌缺血再灌注提供广一个有用的动物模型.
Objective:To develop a new closed-chest model of chronic myocardial ischemia and infarction indogs. Methods:75% and 56% stenosis of left circumflex (LCX) and left anterior descending (LAD) arteries were produced in 15 dogs using stenotic device made of polyvinyl chloride inserted through right femoral artery. Selective coronary arteriography, left cineventriculography and myocardial pathological examination were performed in 9 dogs 5-10 months after operation. Results; Occlusions or 95% stenosis of LCX and 95% stenosis of LAD were found in 5, 3 and 1 dogs respectively by coronary arteriography. Total ejection fraction of left ventricle decreased (P<0. 05) with akinesia in posterobasal and diaphragmatic segments in 5 dogs and hypokinesia in 3 dogs in LCX lesion group. Myocardial fibrosis was significant with calcification, dilatation of T tubule and regional myocardial lysis. Conclusion; This technique provides an useful model for further research on myocardial ischemia and reperfusion.
