精脒对CAOV3卵巢癌细胞端粒酶活性的影响

摘要 目的:探讨精脒对CAOV3卵巢癌细胞的生长和端粒酶活性的影响。方法:应用MTT法观察精脒对CAOV3卵巢癌细胞生长的影响,应用TRAP-银染色法检测精脒对CAOV3卵巢癌细胞端粒酶活性的影响。结果:在细胞培养过程中,外源性增加精脒浓度可使细胞生长速度加快。用不同浓度精脒室温处理的CAOV3细胞端粒酶提取液2小时后,端粒酶活性与对照组差异无显著性。CAOV3细胞经不同浓度精脒处理3天后的端粒酶活性与对照组相比明显增高。结论:精脒浓度的升高可引起端粒酶活性的增高,精脒引起的细胞增殖与端粒酶活性增高有关。 Objective: To explore the effect of spermidine on the cell growth and the telomerase activity in CAOV3 cells. Methods: MTT assay was used to detect the effects of spermidine, and TRAP method was used to detect the telomerase activity in spermidine treating CAOV3 cells.Results:During cell growth, increase exogenous spermidine resulted in the increase rate of cell proliferation. Telomerase activity of CAOV3 cells telomerase extraction treated with spermidine for 2 hours had no significant difference from the control group. Telomerase activity of CAVO3 cells treated with spermidine for 3 days significantly increased compared to the control group. Conclusion: Telomerase activity increased with the increasing of spermidine levels. Spermidine inducing cell proliferation was related to the telomerase activity increasing.
