The mechanisms of central hypersensitivity evoked by glyceryl trinitrate infusion in rats:the involvement of Glu-NMDAR pathways

摘要 【目的】研究硝酸甘油诱导的大鼠偏头痛与脑内兴奋性神经递质谷氨酸(Glu)及其受体N-甲基-D-天门冬氨酸受体(NMDAR)的关系。【方法】静脉注射硝酸甘油构建偏头痛大鼠模型;应用免疫组化技术检测大鼠三叉颈复合体、三叉神经节Glu含量;应用Western-Blot检测三叉颈复合体NMDAR2B亚单位及该亚单位1472位点酪氨酸磷酸化水平。【结果】静脉注射硝酸甘油大鼠脑内三叉神经节Glu含量减少,三叉颈复合体Glu含量增加;Western-Blot表明NMDAR的2B亚单位蛋白表达在三叉颈复合体无明显变化,该亚单位1472位点酪氨酸磷酸化水平明显增高。【结论】神经传导通路兴奋性的改变可能在偏头痛病理生理发生过程中起作用。 [Objective] To observe the expression of glutamate in trigeminocervical complex and trigeminal ganghon of rat following GTN infusion, and to speculate the levels of NR2B, phospho-NR2B protein expression in cell membrane of rat trigeminocervical complex for the investigation of relationships between Glu-NR pathway and the central hypersensitivity evoked by Glyceryl trinitrate infusion in rats. [Methods] Rat migraine model was established with GTN 2μg·(kg·min)^-1 infusion for 30 min. Immunohistochemical staining was used to observe the expression of glutamate in trigeminocervical complex, and in trigeminal ganglion of rat following GTN infusion. The levels of NR2B and phospho-NR2B protein expression in cell membrane of rat trigeminocervical complex were detected following GTN infusion by Western-blot. [Results] The glutamate increased in rat trigeminocervical complex while decreased in trigeminal ganglion. The level of NR2B protein expression remained the same after GTN infusion. The increase of phespho - NR2B protein expression in rat trigeminocervical complex was detected after GTN infusion. [Conclusions] These results demonstrate that the involvement of Glu - NMDAR pathway mediated mechanisms in central hypersensitivity evoked by glyceryl trinitrate infusion in rats.
