三羟异黄酮对去卵巢大鼠主动脉雌激素β受体和原癌基因c-fos表达的影响 被引量:2

摘要 目的观察三羟异黄酮(GST)对去卵巢大鼠主动脉雌激素β受体(ERβ)和原癌基因c-fos表达的影响。方法将成年雌性大鼠40只随机分为4组:假手术对照组(A组);去卵巢组(B组);去卵巢+雌二醇组(C组);去卵巢+GST组(D组)。给药8周后处死大鼠。免疫组化法检测主动脉中ERβ和原癌基因c-fos的表达。结果1)去卵巢手术大鼠主动脉ERβ的表达明显降低,GST与雌二醇明显增加去卵巢大鼠主动脉ERβ的表达。2)去卵巢手术增加大鼠主动脉原癌基因c-fos的表达,但GST与雌二醇能阻止c-fos表达增加。结论GST对血管ERβ的表达具有调节作用;GST可抑制血管平滑肌原癌基因c-fos的表达。这可能是植物雌激素防止动脉粥样硬化,发挥心血管保护作用的机理之一。 Objective To study the effects of genistein (GST) on the expression of estrogen receptor β (ERβ) and the expression of c-fos gene in the aorta of ovariectomized rats. Methods Forty female Wistar rats were randomly divided into four groups: sham-operated (control), ovariectomized (OVX), ovariectomized with 17β-E2 replacement (OVX + E2 ), ovariectomized with genistein replacement ( OVX + GST) group. After 8 weeks' replacement therapy, the rats were sacrificed and the expression of ERβ and c-fos in the aorta was studied by immunohistochemistry. Results Ovariectomy significantly decreased ERβ expression and increased c-fos ex-pression in aorta; while replacement therapy, GST and E2, attenuated the effect of ovariectomy manifested by in-creasing ER expression and decreasing c-fos expression in aorta. Conclusion GST may modulate the expression of ERβ in the arteries of ovariectomized rats and play a beneficial effects on cardiovascular system which was asso-ciated with decreases in expression of c-fos gene in aorta.
