广西何首乌GXHSWAQⅠ在鼻咽癌细胞放射增敏过程中对基因HIF-1α的影响 被引量:3

摘要 目的:探讨广西何首乌中蒽醌类化合物GXHSWAQⅠ,在鼻咽癌CNE-1细胞的放射增敏过程中对乏氧诱导因子表达的影响。方法:MTT比色法确定GXHSWAQⅠ对鼻咽癌CNE-1细胞株体外增敏剂量和放射增敏比,HE染色观察GXHSWAQⅠ放射增敏后细胞形态学的改变,逆转录—聚合酶链反应(RT-PCR)检测GXHSWAQⅠ作用前后,乏氧诱导因子HIF-1α的表达情况。结果:无细胞生长抑制作用浓度的GXHSWAQⅠ低浓度(3.90mg/L)和高浓度(7.80mg/L)组的放射增敏比分别为1.23和1.58,放射和联合用药的细胞发生明显的细胞形态学改变,同时下调基因HIF-1α的表达。结论:GXHSWAQⅠ对鼻咽癌CNE-1细胞具有放射增敏作用,其机制与下调乏氧诱导因子HIF-1α表达有关。 Objective: To investigate the radiosensitiza- tion o{ GXHSWAQ I (an anthraquinone compound extracted from Radix Polygoni Multiflori)in human nasopharyngeal carcinoma cell lines the expression of HIF-1α after GXHSWAQ I treated. Methods: The sensitive enhancement ratio (SER) of GXHSWAQ I on the human CNE-1 cell lines were evaluated by MTT colorimetric assay. Morphological changes was observed by HE staining. HIF-1α gene expression was detected by RT-PCR. Result: At the non-cytotoxic concentration of 3.90 mg/L and 7.80 mg/L GXHSWAQ I could inhibit cells growth post-irradiation. The sensitive enhancement ratio (SER) of GXHSWAQ I were 1.23 and 1.58 respectively. When irradiated by and exposed to GXHSWAQ I ,CNE-1 cells showed morphological changes. The radio sensitivity related gene HIF-1α in cells were down-regulation after treated with different concentration of GXHSWAQ I. Conclusion: GXHSWAQ I has shown a radiosensitizing effect in vitro on nasopharyngeal carcinoma CNE-1 cells. Radiosensitization by GXHSWAQ I is possibly associated with the expression of HIF-1α down-regulation.
