摘要
目的研究间尼索地平(m-Nis)对KCl及ET-1所致离体家兔冠状动脉分支收缩的拮抗作用。方法通过Myograph System-610M描记离体家兔冠状动脉环的张力变化。结果m-Nis和Nis几乎可以完全抑制70 mmol.L-1KCl诱发的离体家兔冠状动脉分支收缩,且均有浓度依赖性,m-Nis的作用比Nis更强;不同浓度的m-Nis(1×10-5、1×10-6、1×10-8、1×10-10mol.L-1)均可以抑制ET-1引起的离体家兔冠状动脉分支收缩;m-Nis使ET-1收缩冠状动脉的效能和效价均降低,随着剂量的增大,抑制作用增强。结论m-Nis对KCl及ET-1诱发的家兔冠状动脉分支收缩均有良好的抑制效果。
OBJECTIVE To study the effects of m- Nisoldipine(m -Nis) on KC1 and ET- 1 induced contractile responses in rabbit coronary artery smooth muscle. METHODS The tension of rabbit coronary artery rings was measured by Myograph System - 610M. RESULTS m - Nis and Nis could completely antagonize the contractile responses of isolated rabbit coronary artery branch induced by 70 mmol· L^-1 KCl in a concentration dependent manner. The antagonistic effect of m - Nis was stronger than that of Nis. Different concentration of m - Nis ( 1 × 10 ^-5,1 × 10^ -6 1× 10 ^-8 1 × 10^ -10 mol· L^ -1 ) could antagonize the contractile responses of isolated rabbit coronary artery branch induced by ET - 1 and reduce both efficacy and potency of ET - 1. CONCLUSION m - Nis antagonizes the contraction responses of rabbit coronary artery smooth muscle induced by KCl and ET - 1.
