摘要
目的观察骨形态发生蛋白-7(BMP-7)在单侧输尿管梗阻(UUO)肾间质纤维化大鼠模型中的表达情况及与肾损伤的关系。方法将雄性Wistar大鼠48只随机分为正常对照组、假手术组、UUO模型组,分别采用免疫组化和原位杂交方法检测肾组织中BMP-7及BMP-7 mRNA的表达和定位。结果与正常对照组和假手术组相比,UUO模型组BMP-7在肾小管上皮细胞和肾间质中表达明显下降,而肾小球中均未见表达;且术后3d BMP-7表达即有下降,随梗阻时间延长,其表达量进行性下降。结论BMP-7在肾间质纤维化早期即出现表达量下降,随病程进展而进行性降低。BMP-7表达下调可能是肾间质纤维化发生发展的机制之一。
Objective The aim of the study was to investigate the expression of bone morphngenic protein-7 ( BMP-7 ) in rat model of renal interstitial fibrosis induced by unilateral ureteral obstruction ( UUO), and its relationship with renal damage. Methods 48 male Wistar rats were randomly allocated into 3 groups, including normal group, sham-operated group and UUO group. The expression and location of BMP-7 and BMP-7 mRNA in renal tissues were examined by immunohistochemistry and hybridization in situ. Results The expression of BMP-7 in renal tubular epithelial cells and interstitial tissue was shown to be lower in the UUO group than in the normal and sham-operated group,thongh it was not found in renal glomerular cells.The level of BMP-7 expression started to decline from the 3^rd day after the operation in UUO group, which was inversely correlated with the duration of obstruction. Conclusion The reduced expression of BMP-7 is observed at the early stage of renal interstitial fibrosis, which is further decreased with the disease progression. The down-regulation of BMP-7 level is suggested to be related to developing renal interstitial fibrosis.
