HPLC法测定生丹颗粒中补骨脂素及异补骨脂素的含量 被引量:5

摘要 目的建立生丹颗粒中补骨脂素、异补骨脂素的含量测定方法。方法采用高效液相色谱法,以AgilentZorbaxSB-C18(4.6mm×250mm,5μm)为色谱柱;流动相:甲醇-水(50∶50);检测波长:246nm;流速:1.0ml/min;柱温:40℃;结果补骨酯素在1.135~45.400μg.ml-1范围内,浓度与峰面积呈良好的线性关系(r=0.9998);异补骨酯素在1.035~41.400μg.ml-1范围内,浓度与峰面积呈良好的线性关系(r=0.9999);样品补骨脂素的平均回收率98.0%,RSD为1.1%(n=6);异补骨脂素的平均回收率97.8%,RSD为0.9%。结论此方法简便、快速,可作为生丹颗粒中补骨脂素、异补骨脂素的含量测定方法。 Aim To establish an HPLC method of determining psoralen and isopsoralen in Shengdan keli. Methods HPLC, Agilent Zorbax SB-C18 (4.6ram × 250mm,5 μm) was used as the chromatographic column, and methanol-water(50: 50) was used as the mobile phase. The detection wavelength was 246nm; the flow rate was 1.0ml/ min; and the column temperature was 40℃. Results The liner range of psoralen was 1. 135 -45. 400μ g·ml^-1 ( r = 0. 999 8), and the liner range of isopsoralen was 1. 035 -41. 400p, g·ml^-1( r =0.999 9). The average recovery of psoralen was 98.0%, RSD = 1.1% ( n =6) ; The average recovery of isopsoralen was 97.8% , RSD =0.9% ( n = 6) . Conclusion The results showed that this method was reliable, accurate and could be used to separate and determine psoralen and the isopsoralen in Shengdan keli.
