摘要 目的:建立一种简便、快速、灵敏、准确地鉴定中药中莨菪碱类成分混入的高效液相色谱-阵列二极管检测器(HPLC- DAD)、高效液相色谱-串联质谱(HPLC-MS)分析方法。方法:色谱柱为Lichrospher Si正相色谱柱(250 mm×4.6 mm,5μm), DAD检测条件:流动相:30 mmol·L-1醋酸铵溶液-乙腈=30:70,检测波长210 nm,流速1.0 ml·min-1。HPLC-MS以离子阱为检测器,采用正离子检测模式进行检测。结果:运用此方法成功的确证中药煎剂中阿托品、山莨菪碱、东莨菪碱类成分。结论:本方法简便准确,可做为中药中莨菪碱类成分混入的检测方法。 Objective: To establish a way to identify the mixture of hypscyamine component into TCM by HPLC-DAD combined with HPLC-MS. Method: Lichrospher Si normal phase chromatographic column (250 mm × 4.6 mm, 5 μm) was used and DAD was taken as the detection; 30 mmol·L^-1 ammonium acetate solution methanol and acetonitrile(30: 70) were used as mobile phase,detection wavelength was at 210 nm and flow rate was 1.0 ml·min^-1. Ion trap was used as the detection in HPLC-MS with the positive ion model. Result: Atropine, Anisodamine and scopolamine were identified successfully in the TCM decoction. Conclusion: The method is easy and reliable, which can be used to detect the mixture of the hypscyamine into the TCM.
