HPLC法测定制何首乌中大黄素的含量

Determination of Emodin in Radix Polygoni Multiflori Preparata by HPLC

目的:建立用HPLC法测定制何首乌中大黄素含量的方法。方法:Kromasil C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-水-冰醋酸(60:40:1);检测波长:437 nm;流速:1.0 ml·min-1;柱温:35℃。结果:大黄素在0.06～0.70μg范围内线性关系良好,r=0.999 9,平均回收率100.5%,RSD为1.6%(n=9)。结论:本方法专属性强,灵敏度高,重现性好,适用于制何首乌的质量控制。

Objective： To establish an HPLC method for determination of emodin in radix polygoni muhiflori preparata. Method： The kromasil C18 column （250 mm ×4.6 mm, 5 μm） was used, the mobile phase was aeetonitrile -water- glacial acetic acid （60： 40：1 ） , the detection wavelength was at 437 nm, the flow rate was 1：0 ml.min-1. The column temperature was 35 ℃. Result： The linear range of emodin was 0.06 - 0.70 μg （ r = 0.999 9 ）. The average recovery was 100.5%, RSD was 1.6% （ n = 9 ）. Conclusion ： The method is specific and sensitive, it can be used for quality control of radix pelygonum multiflori preparata.
