梁金菇多糖对人脐血单个核细胞增殖活性的增强效应及作用机制的实验研究

摘要 [目的]研究梁金菇多糖(LJPS)对人脐血单个核细胞(CBMC)增殖活性和白介素-2受体(IL-2R)生成的影响,为栽培的梁金菇药用价值的开发提供实验依据。[方法]采用四甲基氮唑盐(MTT)比色法检测LJPS体外对CBMC增殖活性的影响,采用碱性磷酸酶-抗碱性磷酸酶(APAAP)桥联酶标法检测LJPS对CBMC表面IL-2R生成的影响。[结果]1)LJPS能够在体外单独或与人重组白介素-2(rIL-2)协同刺激CBMC的增殖(分别为P<0.05和P<0.005),LJPS的最佳浓度为1mg/L。并且LJPS在rIL-2诱导的脐血-淋巴因子激活的杀伤细胞(CB-LAK)早期加入始能明显增强CB-LAK细胞的增殖活性(P<0.01)。2)LJPS单独或与rIL-2协同均能促进CBMC细胞表面IL-2R的表达(分别为P<0.002,P<0.001)。[结论]LJPS对静止的CBMC有活化效应,而且能使rIL-2诱导的CB-LAK有更强的增殖活性,其作用机制与LJPS能够促进CBMC表达IL-2R有关。 [Objective] To study the augmentation effects on the proliferation activity and the expression of interleukin-2 receptor on human umbilical cord blood mononuclear cells(CBMC) activated by LJPS alone or recombinant rIL-2. It could provide the experimental evidence for medical use of LiangJin Mushroom cultivated in China. [Methods] The effects of LJPS on proliferation activity of CBMC have been investigated by MTT colorimetric assay. APAAP enzyme immunoassay was used to study the effects of LJPS on expression of IL-2 receptor on CBMC. [Results] LJPS alone or in combination with rIL-2 can stimulate the proliferation of CBMC in vitro(P〈0.05 and P〈0.005 respectively), the optimum concentration of LJPS was 1 g/L. When rIL-2 and LJPS were put on the culture system at the same time, the pro- liferation activity of CB-LAK cells can be enhanced obviously(P〈0.01). Both LJPS alone or recombinant rIL-2 can strengthen the expression of IL-2 receptor on CBMC(P〈0.002 and P〈0.001 respectively). [Conclusions] LJPS can activate the static CBMC and enhance the proliferation activity of the activated CB-LAK by rIL-2, the mechanism may be related to the enhancing effect of LJPS on the expression of IL-2R on CBMC.
