维生素C防治阿霉素心肌损伤的实验和临床研究 被引量:4

摘要 本文旨在探讨维生素C对受阿霉素损伤的原代培养的大鼠心肌细胞的保护作用及其临床意义。培养3天的心肌细胞用1.8μmol/L的阿霉素处理作为研究模型,然后在培养基中加入不同浓度的维生素C培养4小时。结果观察到合适浓度的维生素C能显著减少受损心肌细胞乳酸脱氢酶(LDH)释放,减少细胞内丙二醛(MDA)的形成,并改善细胞超微结构的变化,显示其具有抑制脂质过氧化,保护心肌细胞的作用。观察了10例接受阿霉素治疗的白血病和淋巴瘤患者,合用维生素C治疗一周后血中超氧化物歧化酶(SOD)和MDA的变化,发现维生素C能显著提高患者的SOD水平。 The aim of this study was to investigate the effects of Vit C on the primary cultured myocardial cells exposed to Adriamycin (ADM) and to evaluate it's clinical significance. The three-day cultured myocardoal cells treated with 1. 8μmol/L ADM were ed as a study medel, then different doses of Vit C (0. 056, 0. 17,0. 56mmol/L) were added to the media incubated for 4 hours. The lactate dehydrogenase(LDH) activity in the media and the content of malondialdehyde (MDA) in the cells were measured, and the ultrasructnd changes of myocytes were also studied. It was shown that the activities of LDH in the media with the addition of Vit C was lower than tha treated with ADM alone (P < 0. 05 ), and the content of MDA in the cells were reduced (P<0. 05). Microscopically, the cells treated with ADM were shown to have the swelling of mitochondria and disruption of critae lysis of myofibrils. After addition of Vit C, the above changes were improved. Clinical study has shown that the SOD level in blood was increased after one week Vit C treatment.
