生物免疫刺激剂对大鼠外周血淋巴细胞和肝大颗粒淋巴细胞生物活性的影响

INFLUENCE OF BIOLOGICAL IMMUNE STIMULANTS ON THE ACTIVITY OF LYMPHOCYTES IN PERIPHERAL BLOOD AND LARGE GRANULAR LYMPHOCYTES IN THE LIVER OF RAT

摘要 正常大鼠在体内应用干扰素(IFNα—2b),卡介苗(BCG)或二者联用后,检测外周血淋巴细胞转化率和酸性非特异性酯酶(ANAE)阳性淋巴细胞数量的变化。同时从正常大鼠肝内分离出大颗粒淋巴细胞(LGL),体外分别受IFNα-2b、BCG或二者联合作用后,以3H-TdR掺入实验测定生物免疫刺激剂(BIS)对LGL抗肝癌细胞作用的影响。结果:(1)单用IFNα-2b对大鼠淋巴细胞母细胞化有一定的抑制作用,抑制率为21.7%;单用BCG使大鼠淋巴细胞转化率提高了27.9%,二者联用则可消除IFNα-2b对大鼠淋巴细胞母细胞化的抑制作用。(2)IFNα-2b、BCG或二者联用均使大鼠外周血ANAE阳性淋巴细胞显著增多,其中BCG作用强于IFNα-2b(P<0.01),以IFNα-2b和BCG二者联用的效果最强。(3)IFNα-2b、BCG均可显著增强大鼠肝LGL的抑肝癌细胞增殖作用,BCG作用较IFNα-2b强,二者分别使肝LGL抑肝癌细胞增殖作用提高了50%和22.4%,IFNα-2b和BCG协同作用的效果最强,使肝LGL的抑肝癌细胞作用提高了63%。实验结果为肝癌免疫治疗的最佳选择提供了充分的科学依据。 After the normal rats were treated respectively with IFNα 2b,BCG or combination of both,the rejuvenation rate of lymphocyte and the number of acid α naphthyl acetate esterase(ANAE)positive lymphocytes in peripheral blood were measured.The large granular lymphocytes(LGL)were isolated from rat liver,and were treated respectively with IFNα 2b,BCG or combination of both in vitro,the influence of biological immune stimulants(BIS)on LGL’s cytostatic activity against human SMMC 7721 hepatoma cells were assayed through measuring 3H TdR incorporation into tumor cell DNA.The results showed that:(1)IFNα 2b reduced the rejuvenation rate of lymphocytes by 21.7%,BCG raised it by 27.9%,the combined use of IFNα 2b and BCG resulted in remaining the rejuvenation rate of lymphocytes in normal level.(2)The number of ANAE positive lymphocytes in peripheral blood increased obviously,the effect of BCG was more effective than IFNα 2b(P<0.01),the combination of IFNα 2b and BCG showed the best result.(3)IFNα 2b and BCG could enhance the cytostatic effect of LGL against hepatoma cell obviously,BCG was more effective than IFNα 2b.IFNα 2b and BCG enhanced the cytostatic effect of LGL against hepatoma cell respectively by 22.4% and 50%.The combined use of IFNα 2b and BCG exhibited the best effect,enhancing the anti hepatoma effect of LGL by 63%.The results provided the most adequate scientific foundation for the best application of hepatoma immunotherapy.
