苦参碱对肝癌患者肿瘤浸润淋巴细胞的影响 被引量:13

摘要 目的:研究不同浓度苦参碱对肝癌患者肿瘤浸润淋巴细胞(TIL)增殖、体外杀瘤活性的影响.方法:将苦参碱分为0.1,0.2,0.4,0.6,0.8g/L5个浓度组,分别加入含有IL-2的DMEM培养基,体外诱导扩增TIL,通过MTT法检测TIL增殖、体外杀瘤活性,采用流式细胞术测定T淋巴细胞亚群,研究苦参碱体外对肿瘤免疫功能的影响.结果:苦参碱低浓度组(终浓度分别为0.1,0.2g/L)对TIL作用不明显,高浓度组(终浓度为0.8g/L)能抑制TIL增殖和体外杀瘤活性(P<0.01),中浓度组(终浓度分别为0.4,0.6g/L)能促进TIL增殖和体外杀瘤活性(P<0.01),CD8+数量有所增加,CD4+/CD8+比值有所下降(P<0.01).结论:合理利用苦参碱可增强患者肿瘤免疫能力. AIM: To explore the effect of different concentra- tions of matrine on proliferation and activity of tumor-infiltrating lymphocytes (TIL) of patients with hepatocellular carcinoma in vitro. METEODS: Different concentrations of matrine (0. 1, 0.2, 0.4, 0.6, 0.8 g/L) were respectively added into DMEM medium supplemented with IL-2. The effect of matrine on proli- feration and activity of TIL in vitro was studied by MTF, and the Phenotype was detected by flow cytometry (FCM). RESULTS: The lower concentration of matrine (0.1,0.2 g/L) had no effect on TIL. High concentration of matrine (0.8 g/L) had a remar- kable inhibitory effect on proliferation and antitumor activity of TIL (P 〈0.01 ). The middle concentration of matrine (0.4, 0.6 g/L) enhanced the proliferation and antitumor activity of TIL in v/tro ( P 〈 0. 01 ). The percentage of CD8^+ increased and the ratio of CD4^+/CD8^+ decreased ( P 〈 0.01 ). CONCLUSION : The reasonable use of matrine could enhance the patients' specific antitumor immune function.
