摘要
目的:探讨血红素氧化酶-1在协调性异种心脏移植中的作用及其机制.方法:建立金黄地鼠对大鼠异种心脏移植模型.随机将实验动物分为4组:对照组、环孢素组、脾切除组、脾切除+环孢素组.观察异种移植物的生存时间.HE染色观察各组移植物组织病理学改变.免疫组化染色观察各实验组不同时间移植物组织中NF-κB,P-selectin,HO-1的表达情况.结果:脾切除组移植物生存时间较对照组和环孢素组延长.脾切除联用环孢素较其它组移植物生存时间延长.移植心脏发生排斥反应时,移植物组织中NF-κB,P-selectin表达呈阳性.长期存活的移植心脏组织中HO-1表达阳性,未见到NF-κB,P-selectin表达.结论:异种移植物血管内皮细胞HO-1表达上调对移植物长期存活有一定意义,对移植物血管内皮细胞具有一定的保护作用.
AIM: To investigate the role of Heme oxygenase-1 (HO-1) in the protection of xenograft endothelium and its mechanism. METHODS: Hamster to rat cardiac transplantation model was constructed. The animals were divided into 4 groups randomly: control group, cyclospofine A (CsA) group, splenectomy group, splenectomy + CsA group. Hamster heart was transplanted into rat abdominal cavity. The survival time of the xenografts were monitored. The histological changes of the xenogafts were investi- gated by HE staining. H0-1, NF-KB and P-selectin expressions were detected immunohistochemically in the xenografts of different groups. RESULTS: The survival time of the xenografts in the splenectomy group was longer than that in the control group and CsA group. The survival time of the xenograft in the splenec- tomy + CsA group was prolonged significantly as compared with the other group. NF-KB and P-selectin expressions were observed in the rejected xenografts. H0-1 expression was positive in the endothelial cells of long-term survived xenografts in the splenecto- my + CsA group. However, no evidence of NF-KB and P-selectin expression was discovered. CONCLUSION: Upregulation of HO-1 can protect endothelium of the xenograft, which is vital to the long-term survival of the xenograft.
