摘要
目的研究抗凋亡基因生存素(survivin)在肝细胞癌中的表达及其与磷酸化蛋白激酶B(Akt)的相关性。方法从30例肝细胞癌组织、11例癌旁组织、11例正常肝组织中提取总RNA及蛋白,利用RT-PCR、蛋白印迹,检测生存素在3种组织中的表达水平;并分析肝癌组织中生存素表达与Akt磷酸化水平的关系。结果肝癌组织中生存素的表达水平明显高于癌旁和正常肝组织(P<0.01);同时低分化、有肿瘤转移的肝癌其生存素表达强度明显高于高分化、无转移的肝癌(P<0.05);相关分析表明,肝细胞癌组织中生存素的表达与Akt磷酸化的表达呈正相关(r=0.8086,P<0.05)。结论Akt磷酸化水平的增高可导致生存素蛋白表达水平的明显增高,生存素、磷酸化Akt异常高表达与肝细胞癌具有密切关系,它们在肝癌的发生、发展中起着重要的作用。
Objective To detect the expression of survivin in hepatocellular carcinoma (HCC) and correlation between expression of survivin and phosphorylation of protrin kinase B(PKB) or Akt in HCC. Methods Western blotting and RT-PCR were employed to test 30 resected HCC and adjacent non-cancerous liver tissues, and normal liver tissues for the expression of survivin. The expression of phospho-Akt was detected by Western blotting. Results The level of mRNA and protein of survivin in HCC was higher than that in adjacent non-cancerous liver tissues and normal liver tissues(P(0.01). The expression of survivin was associated with the differentiation and status of metastasis (P〈0. 05). Westeren blotting revealed a significant positive correlation between survivin and phosphorylation of phospho-Akt ( r= 0. 8086, P 〈 0. 01 ). Conclusion Hyperphosphorylation of PKB/Akt might increase the expression of survivin. Survivin and phosphorylation of phospho-Akt could be of importance for the initiation and development of HCC.
