摘要
为了探讨自发性高血压鼠脑底动脉是否含有内皮素-1(ET-1)能神经纤维分布以及ET-1能神经是否涉及脑血管的神经源性调节及脑血流调节,本文应用免疫组织化学技术观察了自发性高血压鼠和Wistar正常血压鼠脑底动脉(包括大脑前动脉、大脑中动脉、大脑后动脉和基底动脉)ET-1能神经纤维的分布;应用放射免疫法测定自发性高血压鼠脑基底动脉ET-1水平。结果显示:自发性高血压鼠和Wistar正常血压鼠脑底动脉均可见ET-1能免疫反应阳性纤维,似细线状,攀附于血管壁上;高血压鼠ET-1能纤维密度明显高于Wistar正常鼠。此外,高血压鼠脑基底动脉ET-1水平较Wistar正常血压鼠也明显升高。以上结果提示:自发性高血压鼠脑底动脉增加的ET-1能免疫反应阳性纤维可能与脑血管的神经源性调节有关;而升高的ET-1水平可能参与调节高血压时的脑血流量。
The purpose of the present study is to investigate whether endothelin-1 ( ET-1 ) is expressed in cerebral vasculature in spontaneously hypertensive rats (SHR), and whether ET-1 participates in the regulation of cerebral blood flow during high blood pressure. Immunohistochemical technique and radioimmunoassay were used to observe the ET-1 immunoreactive nerves and ET-1 levels, respectively, on cerebral artery, including anterior cerebral artery ( ACA), middle cerebral artery ( MCA), posterior cerebral artery (PCA) and basilar artery (BA). The results showed that: ET-1 is expressed in both cerebral arteries of SHR and normal rats, but the density of ET-1 perivascular nerves was denser in SHR than that of Wistar normotensive rat; The ET-1 levels on basilar artery in SHR were also obviously higher compared with control. These results suggest that the enhanced ET-1 perivascular nerves on cerebral artery may play an important role in the neurogenic regulation of cerebral vasculature in SHR. The possibility that the elevated ET-1 levels of cerebral artery may be involved in the regulation of the cerebral blood flow during pronounced arterial hypertension in SHR is raised.
