摘要 目的研究补肾聪耳胶囊对卡那霉素耳毒性和噪声性内耳损伤的保护作用以及其补肾与抗衰老作用。方法分别采用卡那霉素和噪声损伤制备豚鼠听力损伤模型,老龄大鼠作为老年性聋模型,小鼠作为微循环效应模型,分别给予不同剂量补肾聪耳胶囊,以耳廓反射、脑干诱发电位阈值、耳蜗毛细胞形态变化、甲状腺激素水平及超氧化物歧化酶(SOD)、脂质过氧化物(LPO)为观察指标,比较分析各组的不同变化。结果口服不同剂量补肾聪耳胶囊后,卡那霉素及噪声组耳廓反射、脑干诱发电位阈值、外毛细胞静纤毛缺失率均有显著改善,老年大鼠T4水平升高;高剂量组红细胞、肾及肾上腺SOD活性显著增高,血清LPO水平明显降低。结论补肾聪耳胶囊具有补肾和抗衰老效应,对卡那霉素耳毒性和噪声性内耳损伤具有一定保护作用。 Objective To investigate the protective effects of Bushen Conger Capsules (BSCEC) on animal models with various kinds of inner ear injuries such as toxic agent and noise induced hearing loss, senile and Kidney deficient inner changes and microcirculation disturbance. Methods Hearing loss models of ototoxicity and noise induced injury were prepared by the procedures of Kanamycin injection or noise exposure to guinea pigs, and that with senile and Kidney deficient inner changes were prepared with senile Wistar rats and mice respectively. Then experimental animals were fed with BSCEC at various dosages for a certain length of period. By the end of experiment, comparative analysis was made among different groups of animals to evaluate the changes of relative indicators and their implications in the protective effects of BSCEC on the inner ear injuries by evaluating auricle reflex, brainstem auditory evoked potential threshold, hair cell morphology, plasma thyroid hormone levels, SOD and LPO. Results Following the treatment of BSCEC, auricle reflex, brainstem auditory evoked potential thresholds and outer hair cell damage were significantly improved among the animals with ototoxic and noisy inner injuries, plasma T4 level was markedly elevated among the senile rats, RBC number and the activity of SOD in the tissues of kidney and adrenal gland were obviously increased and the serum levels of LPO was significantly decreased among the animals fed with high dosage of BSCEC. Conclusions Bushen Conger Capsules hold the therapeutic effects on the inner ear injuries, induced by Kanamycin or noise exposure, through the pharmacological effects of Kidney-tonifying and anti-senility.
