β-胡萝卜素和辣椒红素对K562细胞中PPARγ蛋白表达的影响

摘要 为探讨类胡萝卜素能否诱导K562细胞中PPARγ的表达,用β-胡萝卜素和辣椒红素分别作用于K562细胞一定时间后,用MTT法检测其对细胞的抑制率,用流式细胞仪检测处理后细胞的凋亡率,用Western-blotting检测K562细胞中PPARγ蛋白的表达情况。结果显示:β-胡萝卜素对细胞的抑制率呈浓度依赖关系。辣椒红素的作用浓度大于5μmol/L时,对细胞的抑制作用较低浓度时明显增加,并且呈剂量-效应关系。Western-blotting的结果显示一定浓度的β-胡萝卜素和辣椒红素能够增加PPARγ蛋白的表达,进而可能诱导K562细胞的凋亡。 β-Carotene and capsanthin were used to treat K562 cells to investigate whether carotenoids could induce the expression of PPARγin the cell. The inhibition and apoptosis rate of the ceils were detected by means of MTT and the flow cytometry respectively. The expression of PPAR), in K562 cell was examined by Western-blotting. The results showed that the inhibitory rate depended on the concentration of β-carotene. When the concentration of capsanthin was more than 5μmol/L, its inhibiting effect was more obvious than at the low concentration and was dose-dependance. The result of Western-blotting showed that certain concentration of β-carotene and capsanthin can up-regulate the expression of PPAR), and may induce the apoptosis of the cell accordingly.
