摘要
目的探讨大鼠凝血因子ⅦEGF 1片段与脂多糖(LPS)刺激后的大鼠血管内皮细胞(RAOEC)结合能力。方法将0.1mg/L LPS作用RAOEC 2 h、4 h、6 h、18 h,应用免疫荧光法检测TF蛋白在RAOEC的表达,通过荧光显微镜和流式细胞仪检测4μmol/L、8μmol/L、16μmol/L的大鼠凝血因子ⅦE GF 1多肽与LPS作用后的RAOEC的结合能力。结果0.1mg/L LPS能刺激RAOEC表达组织因子(TF),4 h最为明显,4μmol/L、8μmol/L、16μmol/L大鼠凝血因子ⅦEGF 1能结合LPS刺激后的RAOEC。结论大鼠凝血因子ⅦEGF1片段能结合LPS刺激后RAOEC。
Objective To explore the capability of the first epidermal growth factor domain of rat coagulation factor Ⅶ to bind with rate aortic endothelial cell induced by lipopolysaccharide. Methods The expressions of tissue factor on RAOEC induced by 0.1 mg/1 LPS for 2,4,6,18 hours were examined by immunofluorescence method. The capability of binding for EGF 1 with TF were determined by fluorescence microscope and flowcytometry. Results TF is expressed on RAOEC induced by 0.1 mg/1 LPS, the effect is the most remarkable after 4h (P 〈0.01 ),compared with that of control. 4μmol/L,8 μmol/L, 16 μmol/L EGF 1 peptide can bind with the RAOEC induced by LPS. Conclusion The first epidermal growth factor domain of rat coagulation factor Ⅶ to bind with rate aortic endothelial cell induced by lipopolysaccharide.
