摘要
以木香作为实验材料,利用简单提取法、改良CTAB法、改良SDS法、改良CTAB-SDS法和高盐低pH值法对木香基因组DNA进行提取,并对其基因组DNA的纯度和含量进行测定。结果表明:利用以上5种方法提取的木香基因组DNA,在纯度和含量上以改良CTAB法效果最好,其它依次是改良SDS法、简单法、高盐低pH法和改良CTAB-SDS法。
Five methods, including simple extraction method, improved CTAB method, improved SDS method, improved CTAB- SDS method, and method of high salt and low pH value, were used to extract genomic DNA from Rosa banksiae. The purity and quantity of genomic DNA were determined. Result indicates that the improved CTAB method exhibits the highest purity and quantity of genomic DNA, and then followed by improved SDS method, simple extraction method, method of high salt and low pH value, and improved CTAB-SDS method in sequence.
