摘要本文对传统检测基因组DNA甲基化的亚硫酸氢盐方法进行了修改,使它适用于植入前胚胎的DNA甲基化检测研究,从而能够测定少量细胞的(<500个)DNA甲基化状态.结果,用此法成功检测了牛克隆胚胎X染色体中Xist基因的甲基化情况. This paper is a description of a modified protocol that enables detection of DNA methylations in 500 somatic cells and is applicable in animal preimplantation embryos. With this method, the DNA methylation profiles of Xist gene in bovine cloned blastocysts were successfully detected.
