摘要
目的用改良的化学萃取方法,制备去细胞异种神经支架。方法切取青紫蓝兔胫神经,以Triton X-100溶液和脱氧胆酸钠溶液按一定浓度和程序进行化学处理,对萃取神经行组织学HE染色及Laminin和S-100蛋白免疫组织化学观察。结果去细胞异种神经支架的延展性及韧弹性良好,细胞和髓鞘被彻底清除,该神经支架保持了网管柱状组织结构,保留了雪旺细胞(SC)基底膜及其Laminin、神经内膜、神经束膜和神经外膜。结论该方法可为异种神经移植提供较为理想的支架材料。
Objective To prepare the xenogeneic acellular nerve scaffold used a modified chemical extraction method.Methods Sectioning the rabbit tibial nerve and chemically processing it with solution of Triton X-100 and sodium deoxycholate in appropriate density and procedure. Then the extraction nerve segments were observed in morphological and immunohistochemical way. Results The xenogeneic acellular nerve scaffold had a good elasticity and ductility. The basal lamina tube, endoneurium, perineurium and epineurium remain intact and a acellular nerve scaffold was obtained. Conclusion The method can prepare a ideal scaffold material for the graft of xenogeneic acellular nerve.
