摘要 目的寻找高低转移能力肺癌细胞中潜在的蛋白标记以更好的解释肺腺癌转移机制。方法用双向电泳的方法对高低转移能力肺腺癌细胞系AGZY83-a和Anip973进行了比较蛋白质组学分析。选取差异较大的候选差异表达蛋白进行基质辅助激光解析/电离飞行时间质谱仪(MALDI—TOF—MS)分析并鉴定。结果双向电泳胶图分析结果说明两细胞系的蛋白表达差异具有统计学意义。MALDI—TOF—MS鉴定证实这3个蛋白分别为Stathmin 1蛋白、磷酸甘油酸变位酶1(PGAM1)和热休克蛋白27(HSP27)。结论在高转移细胞系Anip973中,HSP表达显著升高,Stathmin 1和HSP27表达显著下降,这3种蛋白与肿瘤的发生和恶性程度密切相关。 Objective To identify potential protein markers between two lung adenocarcinoma cell lines with different metastatic potentials and better understanding the mechanism underlying in the metastasis of lung adenocareinomas. Methods Differential proteome analysis on two lung adenoearcinoma cell lines with high and low metastatic potentials, AGZY83-a and Anip973, was conducted using two-dimensional gel electrophoresis for quantitative differences in protein expression. The Candidate proteins were identified using matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Results Image analysis of the gels revealed that 3 protein spots showed significant differential expression in the two cell lines. They are Stathmin 1, phosphoglycerate mutase 1 (PGAM I ) and HSP27. Conclusion The expression level of HSP27 was elevated in Anip973. However, Stathmin 1 and PGAM 1 were down-regulated in it. It indicates that these proteins associate with tumorigeness and malignant degree.
