摘要
应用ABC酶标法,以菜豆凝集素和花生凝集素(PHA和PNA)作为糖蛋白的分子探针,对胃粘膜细胞癌变过程中糖蛋白的变化进行了回顾性的动态观察,探讨胃癌的发生发展规律。结果显示:胃癌组PHA和PNA受体表达,无论是癌变前还是癌变后均明显高于对照组。胃癌组癌变前后的PHA和PNA受体表达也明显不同,癌变后明显高于癌变前;而对照组前后两次PHA和PNA受体则均无差异。提示胃粘膜细胞癌变过程中伴随有糖蛋白质与量的改变,这种先于形态学变化的分子水平的改变对判断胃癌前病变和胃癌的早期诊断有着重要意义。
In order to determine the relationship between the variety of glycoprotein in gastric mucosa and the development of gastric cancer,a retrospective dynamic observation on the expression of glycoprotein was performed by ABC immunohistochemistry, using phaseolus vulgaris agglutinin (PHA)and peanut agglutinin (PNA) as a molecular probe for discussion on the rules of ocurrence and development of stomach cancer. The results showed that the expression rate of PHA and PNA receptors in cancer group (both before and after canceration of gastric mucosa) was significantly higher than that in control group( P <0.05). In cancer group,obviously different expressions of PHA and PNA were found before and after canceration of gastric mucosa, overexpressions of PHA and PNA receptors were seen after cancer development ( P <,0.05), but no difference was found in control group( P >0.05).These results indicate that the quality and quantity of glycoprotein vary in the process of canceration of gastric mucosa. The overexpressions of PHA and PNA receptors in molecular level are often preceding to morphology change. Therefore, PHA and PNA may act as a following index for identifying real precancerous lesion and judging the diagnosis of gastric cancer in early stage.
