摘要
目的制备乙型脑炎病毒(JEV)与黄热病病毒(YFV)联合诊断基因芯片。方法基于早期实验室研究挑选出28条寡核苷酸探针制备检测芯片。克隆于质粒上的乙型脑炎病毒和黄热病病毒基因片段用于检测探针特异性,经限制性显示技术扩增并标记,与芯片杂交后完成扫描和数据分析。结果JEV、YFV探针与相应的荧光标记样本杂交后,均能检测出阳性荧光信号,而空白对照则检出阴性信号。结论基因芯片技术为病毒检测提供了一种早期、可靠的方法,具有应用于多病毒临床鉴别诊断的前景。
[Objective] To fabricate miemarray for detection of Japanese encephalitis virus and yellow fever virus. [Methods] Based on the early research of 28 probs selected. The specificity of probes was tested by plasmids of the target fragments. Restriction display technology was used to amplify and label the samples. After hybridization, miemarrays were scanned and analyzed. [Result] The result showed that JEVfYFV probes were detected positive signals when hybridizing with corresponding samples. Meanwhile, the blank controls showed negative signals. [ Conclusion] Miemarray technology provides an early and credible method, which may have potential aplieation for clinical multi-viral detection.
