摘要
目的探讨EB病毒(EBV)阳性系统性红斑狼疮(SLE)患者EBV核抗原-2(EBNA2)表达与抗Sm抗体的相关性。方法SLE患者44例,正常人43名作为对照。①采用PCR-Southern杂交技术检测两组外周血单个核细胞中EBV特异性DNA片段,EBV阳性患者进行RT-PCR和Southern杂交检测EBNA2的表达。②ELISA法检测EBV特异性IgM抗体。③免疫印迹法检测血清可提取核抗原(ENA)抗体。结果①SLE组32例EBV阳性,其中EBNA2mRNA阳性20例(活动期9例,稳定期11例),EBNA2mRNA阳性率在活动期和稳定期患者差异无显著性(P>0.05)。②SLE组EBV特异性IgM抗体阳性率为18.18%(8/44),对照组均为阴性,两组有显著性差异(P<0.05)。③SLE抗Sm抗体检出率与EBNA2 mRNA检出率呈正相关(r=0.4107,P<0.05)。结论EBNA2 mRNA表达可能在一定程度上参与SLE的发病和病情进展。
Objective To explore the relationship between the expression of Epstein-Barr virus(EBV) nuclear antigen 2 (EBNA2) and the anti-Sin antibodies in EBV positive SLE patients. Methods EBV specific DNA fragment was tested by PCRSouthern blotting in peripheral blood monocytes of SLE patients(n = 44) and normal controls(n = 43). The expression of EBNA2 in EBV positive samples was detected by RT-PCR and Southern blotting. In SLE patients and normal individuals, the specific EBV IgM antibodies were tested by ELISA. ENA antibodies were tested by Southern-blotting in SLE patients and normal serums. Results Thirty-two cases were positive for EBV in SLE patients, and EBNA2 mRNA was detected in 20 cases of 32 EBV-positive SLE patients (9 were active and 11 were stable). There was no significant difference between the active and stable groups (P〉0. 05). In SLE patients, the positive rate of EBV specific IgM antibody was 18. 18% (8/44), and all the normal individuals were negative (P〈0. 05). There was a significant correlation between the detectable rate of anti-Sin antibody and EBNA2 mRNA in SLE patients (r= 0. 4107, P〈0. 05). Conclusion The expression of EBNA2 mRNA may play a key role in the pathogenesis and progress of SLE.
