摘要
目的探讨健择(盐酸吉西他滨)诱导胰腺癌细胞凋亡可能的途径。方法采用RT-PCR、Western-blot及激光密度扫描技术、AnnexinⅤ-FITC/PI双染法对健择诱导胰腺癌Panc-1细胞发生凋亡过程中caspase-3、bax和survivin基因表达及细胞凋亡率进行了检测分析。结果健择可以诱导胰腺癌细胞发生凋亡,5mg/L组24h凋亡率达(18.83±0.78)%,凋亡率随健择浓度升高而升高,25mg/L组在用药48h后凋亡率高达(42.47±2.77)%。并且caspase-3、bax基因表达量随细胞凋亡的发生而升高,与健择的浓度成正比关系;但survivin基因表达量随细胞凋亡的发生而降低,与健择的浓度成反比关系。结论凋亡调节基因bax、caspase-3的上调及survivin的下调可能与健择诱导胰腺癌细胞凋亡有关系。
Objective To study the role of gemcitabine in the expressions of caspase-3, bax and survivin gene in human pancreatic cancer panc-1 cells apoptosis. Methods The expressions of bax. caspase-3 and survivin gene in pane-1 cells apoptosis induced by gemcitabine were determined by using RT-PCR. Western-blot method and analyzed by laser densitomter. Apoptosis rate was determined by AnnexinV-FITC/PI dyeing. Results Gemcitabine can induce the cells apoptosis of pane-1 cells, and the results show with the occurrence of apoptosis of pane-1 cells, the expression levels of caspase-3 and bax ascended, but the expression level of survivin descended. Conclusion Expressions of bax. caspase-3 and survivin play important roles in the pancreatic cancer cells apoptosis induced by gemcitabine, which show gemcitabine maybe have an important function on the pancreatic cancer treatment.
