^(131)I-D_2C_5在荷瘤裸鼠体内分布实验

摘要 目的探讨131I-D2C5在荷瘤裸鼠体内的生物分布。方法12只荷瘤裸鼠随机分为二组(每组各6只),鼠尾静脉注射131I-D2C5或131I-mIgG4h、24h、48h后,采用γ计数器检测体内放射性分布。结果静脉注射131I-D2C5后4h,131I-D2C5主要分布在血、心、肝、肾、脾中,肿瘤在4h开始略有浓集。24h后肿瘤组织内131I-D2C5聚集达到高峰,此时每克组织的放射性占注射剂量的百分比(%ID/g)达到4.12。131I-D2C5注射后4h、24h、48h肿瘤/肌肉的放射性比值依次为3.96、7.63、8.21,而131I?mIgG依次为3.30、2.80、2.60,两者变化趋势差异显著。结论131I-D2C5能在裸鼠肿瘤组织内特异性聚集,有望用于肿瘤组织的受体显像和导向治疗。 Objective To investigate the biodistribution of ^(131)I-D2C5 in nude mice bearing CNE2 tumor. Methods Twelve nude mice bearing CNE2 subcutaneous tumors were randomized into 2 groups (n = 6) and received the injection of ^(131)I-D2C5 and ^131 I-mIgG via caudal vein, The biodistribution of ^(131)I-D2C5 and ^131 I-mIgG at 4 h, 24 h, 48 h post injection were investigated by γ counter technologies. Results Four hours after intravenous injection,^(131)I-D2C5 was mainly distributed in blood, heart, liver, kidneys and spleen, and concentrated in the tumors slightly. The distributions of ^(131)I-D2C5 in tumors reached the peak at 24 h post injection when %ID/g was 4. 12. The ratio of tumor/muscle at 4 h, 24 h, 48 h post injection was 3.96, 7. 63, 8. 21 respectively, while the ratio of tumor uptake at 4 h, 24 h, 48 h with ^131I - mIgG was 3, 30, 2. 80, 2. 60. There was significant difference between the two groups. Conclusion ^(131)I-D2C5 was specifically localized in tumor lesions of nude mice and may be useful in tumor detection and targeted therapy.
