摘要
[目的]为了找到适合原球茎增殖的培养条件,为工厂化育苗提供技术支撑。[方法]以铁皮石斛茎段诱导出的原球茎为实验材料,研究了不同基本培养基、不同碳源、不同浓度的脱落酸(ABA)和培养方式对其增殖的影响。[结果]在5种培养基中,原球茎增殖倍数均呈上升的趋势,其中改良MS的增殖倍数明显高于其他培养基;在低浓度的条件下,随ABA浓度的增加,原球茎的增殖速度加快,当浓度为0.5 mg/L时,增殖最大;蔗糖为碳源时原球茎增殖的效果比葡萄糖、果糖好;固体培养和液体振荡培养时原球茎增殖倍数最高。[结论]在改良MS中加入30 mg/L蔗糖和0.5 mg/L ABA,进行固体培养或液体振荡培养,铁皮石斛原球茎增殖的效果好。
[Objective] The objective was to discover the suitable culture condition for protoeorm muhiplieation and provide technique support for industrial seedlhlg-raising. [Method] With the protocorm induced from the sterns of Dendrobium candidwn Wall.ex Lindl as the tested materials, the effects of different basic medium, carbon source, ABA concentration and culture ways on the protoconn multiplication were studied. [ Result ]Among 5 kinds of medium, the proliferation multiple of protoconn had rising trend, and that in improved medium was higher than that in other medium. Under low concentration, as the ABA concentration increased, the proliferation speed of protoconn was quickened, with most proliferation at 0.5 mg/L. When cane sugar was as carbon source, the proliferation effect was better than that with glucose and fructose. The proliferation multiple was highest in solid culture or in liquid shake culture. [Conclusion] The protoconn multiplication of D. candidurn could got good effect in solid culture or in liquid shake culture with the improved MS medium 'added 30 mg/L cane sugar and 0.5 mg/L ABA.
