三倍体毛白杨组培快繁技术试验研究 被引量:2

摘要 分别以三倍体毛白杨植株腋芽(或顶芽)、嫩叶片、嫩茎段等为材料,进行愈伤组织诱导、芽分化、生根等试验,探讨三倍体毛白杨组培快繁技术。结果表明:①以自然状态植株腋芽(或顶芽)、嫩叶片以及无菌苗叶片在适宜的培养基中,均可产生质量较好的愈伤组织,其中以腋芽(或顶芽)产生愈伤组织的频率最高,达100%;以无菌苗叶片为材料,愈伤组织诱导率为53.3%-80.0%。②以愈伤组织为材料,不定芽分化频率达13.3%-26.7%;无菌苗叶片经20天培养后,每叶片可产生不定芽5-8个;自然状态下嫩茎段在普通培养室与人工气候箱中均可培养出芽,其中前者芽体小、数量多,后者芽体壮而高、数量少。③在1/2MS+NAA0.2mg/L,1/2MS+IAA0.2mg/L,1/2MS+IBA0.2mg/L三个培养基均可诱导生根,生根率均在70.0%以上,其中以1/2MS+NAA0.2mg/L效果最佳。④黄壤土+砻糠灰+河沙按2:1:1配比(体积比)作为三倍体毛白杨组培苗移栽基质,成活率可达92.4%,且生长性状优良。 Taking sprouts,tender leaves,tender stem-sections of triploid Populus tomentosa Carr. for experiment samples respectively,experiments such as callus formation,buds differentiation,roofing were conducted.The results showed as follows:The fast,sprouts and tender leaves at natural state and leaves of non-bacteria seedlings were taken explants,quantitive callus could be produced in proper mediums among which production rate in sprouts was the highest,amounting to 100% ,and that in non-bacteria seedlings for 53.3%-80.0%. The second,callus were taken samples,differentiation rate of adventitious buds amounted to 13.3%-26.7%;5-8 buds were produced on a piece of leave of non-bacteria seedlings alter 20d's culturing; tender stem-sections at natural state were taken samples,buds could be produced in common culture room and artificial climate room, but the former small and numberal,the latter high and little. The third,rooting rate of tube-seedlings amounted to up to 70% on three mediums including 1/2MS+NAA0.2mg/L,1/2MS+IAA0.2mg/L,1/2MS+IBA0.2mg/L among which the former was the best. The forth,tube-seedlings grew well and the transplantation living rate amounted to 92.4% on the ground substance composed of yellow soil, 'long kang'ash and river sand(2 : 1 : 1 by volume).
