摘要 目的检测兔眼结膜上皮细胞对5’-溴脱氧尿嘧啶(BrdU)的标记保留情况,为进一步探索结膜上皮干细胞的定位提供依据。方法实验1组为短期标记保留实验,采用一次性兔腹腔注射BrdU50mg/kg,不同时间间隔免疫组织化学法分析兔眼各部位结膜组织BrdU标记保留细胞。实验2组为长期标记保留实验,连续7d每日注射BrdU50mg/kg,观察6周后兔结膜上皮的BrdU标记阳性细胞。结果实验1组于1、3、5、7d,BrdU标记阳性细胞数量的高峰位置从睑皮肤黏膜交界处向穹隆部移行,所有切片的睑皮肤黏膜交界处均可见较多的BrdU标记阳性细胞。实验2组的结膜上皮BrdU标记阳性细胞主要保留在睑皮肤黏膜交界处。结论短期和长期标记保留实验说明兔眼睑皮肤黏膜交界处可能含有兔结膜上皮干细胞。 Objective The conjunctival epithelium must be replaced from locally concentrated or randomly distributed foci of stem cells. As the recent studies, the region where conjunctival epithelial stem cells stay has not been determined. In this study, the label-retaining cells of rabbit conjunctival epithelium with bromodeoxyuridine(BrdU) were detected in order to provide the basis for location of conjunctival epithelial stem cells. Methods Fifteen New Zealand white rabbits were divided into two groups randomly. In group 1 , 12 rabbits were injected intraperitoneally with BrdU 50 mg/kg once, and then the label-retaining cells of rabbit ocular epithelium were detected after 1 day, 3,5 and 7 days. In group 2, BrdU 50 mg/( kg. d)were injected intraperitoneally for 7 days. The label-retaining cells of ocular epithelium of rabbit were detected after 6 weeks. Results In test group 1 ,rabbits were killed after 1 day, 3,5 and 7 days, the peak position of numbers of label-retaining cells migrated from mucocutaneous to fornix. A focus of BrdU-labeled conjunctival cells remained in the mucocutaneous junction at various time points. In group 2, BrdU-labeled nuclei were retained at the mucocutaneous junction. Conclusion The conjunctival epithelial stem cells of rabbit may locate in mucocutaneous junction in both short-term and long-term label-retaining experiments.
