力竭性游泳对大鼠胰岛 A、B 细胞形态学变化的影响——免疫组化及原位分子杂交结果分析 被引量:3

摘要 采用免疫组化及原位分子杂交技术,本文研究了负重力竭性游泳对大鼠胰岛A、B细胞分泌功能的影响。主要结果发现:力竭状态下,①大鼠胰岛Ins-IR细胞呈色反应强度增加,而Glu-IR细胞呈色反应强度降低,说明B细胞Ins释放被抑制,A细胞Glu释放增多。②大鼠胰岛B细胞内Pro-InsmRNA杂交信号明显减弱,说明Pro-Ins-mRNA转录受到抑制,Ins合成受抑。③Ins-IR和IAPP-IR细胞未呈现相同程度的呈色反应变化,提示B细胞内存在着含IAPP/Ins不同比率的分泌颗粒,可随机体的机能状态需要而分别释放。 With immunohistochemical PAP method and technique of hybridization in situ,this study investigated the effects of exhaustive swimming on secretion function of islet A and B cells in mice.Results showed that:in exhausted mice,1,color reaction intensity of islet Ins IR cells enhanced and Glu IR cells reduced as compared with controls,suggesting the suppression of B cell releasing and increase of A cell secretion.2,Pro-Ins mRNA hybrid signal significantly weakened in B cells showing inhibition of Pro-Ins mRNA transcription and Ins synthesis 3,IAPP IR cells did not show the same color reaction intensity as Ins-IR cell,indicationg that IAPP was not secreted as equal as Ins.It suggested that the ratio of IAPP/Ins grains in B cells was different and the releases of IAPP and Ins may have seperate way according to the demand of the body.
