FWM+NCR对大隐静脉血管桥内皮源性超级化因子介导血管舒张功能的保护作用

摘要 目的:探讨常温下阜外改良(Fu Wai Modified,FWM)器官保护液+尼可地尔(NCR)对大隐静脉血管桥血管内皮超极化因子(EDHF)介导血管舒张功能的影响。方法:采用器官槽法研究在37℃有氧条件下用重碳酸盐缓冲液(KH)、含0.1 mmol/L NCR的KH液、FWM液、含0.1 mmol/L NCR的FWM液中浸泡血管环1 h后,再用7μmol/L吲哚美辛(Indo)和300μmol/L N-硝基-L-精氨酸(LNNA)作用后,检测30 nmol/L U46619及不同浓度A23187引发的血管收缩舒张反应。结果:光镜下各组未见明显损伤;电镜下含NCR+KH组、FWM组对内皮损伤轻,NCR+FWM组对内皮损伤最轻,KH组损伤重;A23187(10-6mol/L)引发的血管最大舒张反应程度,FWM组[(72.53±10.36)%]、NCR+KH组[(70.85±10.02)%]、NCR+FWM组[(84.55±10.54)%]高于KH组[(49.32±12.48)%,P<0.05];FWM组[(72.53±10.36)%]、NCR+KH组[(70.85±10.02)%]低于NCR+FWM组[(84.55±10.54)%,P<0.05]。结论:FWM液+NCR能很好的舒张血管环,并对血管内皮超极化因子(EDHF)介导血管舒张功能有保护作用。 Objective:To discuss the protective effects of Fuwai modifed solution (FWM) combined with nicoranil (NCR) on the endothelium-derived hyperpolarizing factor (EDHF)-mediated relazation relaxation of saphena vessel graft (SVG). Methods:Fresh saphena rings were sudied in organ chamber after incubating with Krebs-Henseleit bicarbonate solutions (KH), FWM, KH with 0. lmmol/L NCR, FWM with 0. 1 mmol/L NCR at 37 C for 1 hour. Precontraction induced by prostaglandin F2(U46619 30 nmol/L)and endothelium-dependent relaxation induced by calcium ionophore (A23187 10^-10~10^-6mol/L) were measured in the presence of indomethacin (7μmol/L) and LNNA (300μmol/L). Re-sult :No significant changes were found in the 4 groups under the light microscope. Under the transmission eletronmicro-graph, the endothelial cell had a slightly injury in the group NCR+KH and FWM, while group FWM+NCR had a bet- ter protection for endothelia cell than the other three groups. After exposing to FWM+NCR and KH+NCR solution, the maximal relaxation response induced by A23187 (10^-6 mmol/L) was significantly higher than that of in the FWM and KH [(84.55±10.54)%, (70.85±10.02)%, (72.53±10.36)%, (49.32±12.48)%,P〈0.05 respectively]. Conclu-sions:FWM solution combined with NCR have a better preservation effect on the EDHF-mediated relaxation of SVG.
