结直肠癌组织中ECM1基因水平的测定及临床意义 被引量:7

摘要 目的:探讨细胞外基质蛋白1基因在结直肠癌中的表达及其意义.方法:基于TaqMan-MGB荧光探针技术,建立实时荧光定量RT-PCR方法,检测正常结直肠黏膜组织(n=46),结直肠腺瘤(n=18),结直肠癌(淋巴结未转移)(n=25)和结直肠癌(淋巴结转移)(n=21)中ECM1基因的表达.结果:正常黏膜,腺瘤组织,结直肠癌(淋巴结未转移)组织和结直肠癌(淋巴结转移)组织中ECM1基因的表达均值分别为(9.81±3.16)×10~8拷贝/g RNA,(10.1±3.65)×10~8拷贝/g RNA,(6.89±2.96)×10~9拷贝/g RNA,(5.01±2.22)×10^(10)拷贝/g RNA.正常黏膜和腺瘤组织组间无明显差异(P>0.05);结直肠癌(淋巴结未转移)组明显高于正常黏膜和腺瘤组织组P<0.01);结直肠癌(淋巴结转移)组明显高于其他三组(P<0.01).结论:ECM1基因在结直肠癌中表达升高,并且与结直肠癌的转移相关. AIM: To investigate the expression and significance of the ECM1 gene in colorectal cancer tissue. METHODS: Based on TaqMan-MGB methodology, real time quantitative polymerase chain re- action (RQ-PCR) was used to detect the mRNA levels of ECM1 in tissue from normal controls (n = 46), colorectal adenoma (n = 18), colorectal cancer without lymphatic metastasis (n = 25) and colorectal cancer with lymphatic metastasis (n = 21). RESULTS: The mRNA expression of ECM1 in colorectal cancer tissue including those with and without lymphatic metastasis was significantly higher than that in normal and adenoma tissue [(6.89 ± 2.96) ×10^9 copy/g RNA, (5.01 ± 2.22) ×10^10 copy/g RNA vs (9.81 ± 3.16) ×10^8 copy/g RNA, (10.1 ± 3.65)×10^8 copy/g RNA, P 〈 0.01]. There was no significant difference between normal and adenoma tissue (P 〉 0.05). ECM1 mRNA expression in metastatic colorectal cancer was higher than that in the other three groups (P 〈 0.01). CONCLUSION: Over-expressed mRNA of ECM1 in colorectal cancer tissue correlates with the properties of metastasis in colorectal cancer.
