联合显微切割及PCR方法对结外NK/T细胞淋巴瘤中EB病毒的检测

Combine hand-made microdissection with polymerase chain reaction to detect EBV in nasal NK/T-cell lymphoma

目的:研究结外鼻型NK/T细胞淋巴瘤中EB病毒基因的表达。方法:筛选NK/T细胞淋巴瘤56例,应用手工显微切割进行组织纯化后,用PCR检测EBV的基因序列,并同时对相应组织用EBV抗体做免疫组织化学染色检查,对两种方法进行了比较。结果:56例NK/T细胞淋巴瘤应用手工显微切割进行组织纯化后,用PCR方法EBV阳性率为39/56(69.64%),而用免疫组化方法阳性率为7/56(12.50%),二者检出率有显著差异。结论:显微切割联合PCR技术能明显提高EB病毒检出率和准确性,是检测EBV的有效方法。

Obiective： To detect the expression of Epstein Barr Virus （EBV） in nasal NK/T-cell lymphoma. Methods： According to HE shape and immunohistochemical staining, 56 cases were hand-made microdissection ,then ,were examined for the presence of EBV specific DNA sequence by polymerase chain reaction （PCR）. To contrast,immunohistochemical staining was used to examine the expression of EBV. Results： NK/T-cell lymphoma were positive for EBV-DNA in 39/56 （69.64%）cases. To compare with immunohistochemical staining, EBV-DNA have obviously positive rate in NK/T-cell lymphoma. Conclusion： To compare with immunohistochemical staining, EBV-DNA had obviously positive rate in NK/T-cell lymphoma. Microdissection is very helpful to improve detection rate and veracity.
