神经生长因子 cDNA 探针的制备及应用

摘要 人重组βNGF/pUC19,经HindⅢ/BamHⅠ双酶切后,获βNGFDNA片段(360bp)。用随机引物标记法进行Digoxigenin标记,在小鼠颌下腺原位杂交检测NGFmRNA显示出很高的特异性,该探针的制备为NGF在神经及非神经系统的研究提供新的方法。 Recombinant human-β NGF DNA/pUC 19 was digested with Hind Ⅲ and BamHⅠ and h-β NGF DNA fragments were further purified by lowmelting agrose electrophoresis. H-β NGF DNA was labeled with random priming and then used to demonstrate NGF mRNA at the mouse submandibular glands. It showed the high specificity and sensitivity, easy and rapidity. The Dig-h β NGF DNA probes will supply a new method in researching nerve and nonnerve systems.
