摘要
目的:观察辛基酚(OP)对乳腺癌MDA-MB-231细胞凋亡的影响。方法:以流式细胞分析、免疫细胞化学和RT-PCR等方法观察OP对MDA-MB-231细胞凋亡、bcl2、bax和caspase3表达的影响。结果:4μmol/L、8μmol/L-OP作用MDA-MB-231细胞48 h,其凋亡率分别为26.93±6.76、42.48±8.21(对照4.05±1.14);bcl2表达量荧光值分别为78.05±9.47、42.74±6.49(对照103.12±11.26)。OP降低细胞中bcl2 mRNA表达,增加caspase3 mRNA及其蛋白的表达。结论:OP能诱导MDA-MB-231细胞凋亡,其机制可能是通过上调细胞中caspase3的表达,下调bcl2的表达来诱导细胞凋亡。
Objective: To investigate the effect of octylphenol on the apoptosis of MDA - MB - 231 breast cancer cell. MethOds: Flow eytometry, immunoeytochemistry and RT- PCR were used to observe the apoptosis, gene expression of bcl2, bax and caspase3, protein expression of bcl2 and caspase3 in MDA - MB - 231 breast cancer ceils treated by 4μ mol/L and 8μ mol/L OP rseperately. Results: The apoptosis of MDA -MB -231 breast cancer cells was 26.93 ±6.76 and 42.48 ±8.21 respectively (4.05 ± 1.14 in control) after treated by OP for48 h. The fluorescence value of bcl2 expression was 78.05±9.47 and 42.74 ±6.49 respectively ( 103. 12 ± 11.26 in control). OP decreased the mRNA level of bcl2, increased the mRNA and protein expression of caspase3, but didnt increase the mRNA level of bax significantly. Conclusion: OP can induce apoptosis of MDA- MB-231 breast cancer cells. The mechanism may be related to the up - regulation of caspase3 expression and down - regulation bcl2 expression.
