鼻咽癌放射敏感性异质性的形态学差异观察 被引量:3

摘要 目的探讨鼻咽癌放射敏感性异质性与形态学变化的相关性。方法应用电镜观察并用体视学技术测算,比较鼻咽癌细胞系CNE-2Z亚克隆株H5和S1,于体内体外照射后形态学改变的差异;用流式细胞仪、荧光显微镜检测H5和S1放射后凋亡率的不同,用Western-blot检测H5和S1放射后凋亡蛋白Bcl-2表达的差异,以期探讨其形态学异质性机制。结果H5和S1照射后形态学改变差异有显著性;照射后的H5比S1凋亡率高,对射线敏感;S1细胞的各个照射剂量组Bcl-2蛋白表达无明显差异;而H5细胞的各个照射剂量组Bcl-2蛋白表达随照射剂量的增加而递减。结论H5、S1照射后形态学的差异变化可用于预测其放射敏感性的异质性。 Objective To study the correlation of radiosensitive heterogeneity with morphological differences in nasopharyngeal cancer (NPC). Methods The subtypes H5 and S1 of NPC cell line CNE-2Z were irradiated in vivo and in vitro. The morphological changes before and after irradiation were observed with electron microscope and stereological measurement; the apoptosis-inducing capacity of H5 and S1 was detected by FCM and fluorescence microscope; Bcl-2 protein expression of H5 and S1 cells was detected by Western-blot. Results The morphological changes in H5 and S1 after irradiation had statistical difference; H5 had a higher apoptosis rate and was more radiosensitive than S1. Conclusion Changes in the morphological difference between H5 and S1 after irradiation can be used to predict their radiosensitive heterogeneity.
