摘要
目的探讨肝癌及癌旁组织中P-Smad2蛋白与PPM1A的表达及意义。方法采用免疫组织化学技术检测31份肝癌(HCC)、25份癌旁组织及13份非癌性肝组织中P-Smad2蛋白和PPM1A的表达。结果①癌旁和病理分级小于Ⅱ级的癌组织中PPM1A表达以核为主,胞浆表达弱或不表达,病理分级大于Ⅲ级及以上的癌细胞中则以胞浆表达为主,正常、慢性肝炎和肝硬化组织中,PPM1A表达以核为主,胞浆无表达,差别有统计学意义(P<0.05)。②P-Smad2在正常肝细胞、慢性肝炎、肝硬化、癌旁和病理分级Ⅰ-Ⅱ级的癌组织中表达以胞核和胞浆明显,胞浆表达主要聚集在核周,病理分级分级Ⅱ-Ⅲ级及以上的癌组织中,P-Smad2表达则以核为主,差别有显著性(P<0.05)。③PPM1A和P-Smad2在癌组织的表达部位呈现负相关性(r=-0.345,P=0.001)。结论PPM1A与P-Smad2在肝癌组织的胞核和胞浆表达转位及强度与其组织病理改变有密切关系,二者可能通过其相互作用,共同参与肝癌的发生发展机制。
Objectives To explore the expression of phosphorlated Smad2 (P-smad2) and magnesium-dependent protein phosphatase 1 alpha (PPM1A) in hepatocellular carcinoma (HCC) and adjacent liver tissues. Methods Expression of P-smad2 and PPM1A was detected by Envision immunohistochemical technique in 31 cases of HCC, 25 cases of adjacent tissues and 13 cases non-tumor liver tissues. Results ① Expression of PPM1A mainly was in the nucleus of Edmondson-Steiner pathologic grades Ⅰ and Ⅰ - Ⅱ HCC, the adjacent and other non-HCC tissues, but in the cytoplasm of grade Ⅱ-Ⅲ HCC. So the location of PPM1A in HCC was positively correlated with the progress of the tumor grade. ② P-smad2 was expressed in the nucleus and cytoplasm of liver cells. The positive rate and expression intensity of P-smad2 in the nucleus of HCC were the same as those in the adjacent liver tissues, but their locations were different. Most of P- Smad2 was in the nucleus and cytoplasm of grade Ⅰ and Ⅰ - Ⅱ HCC and the adjacent tissues, but only in the nucleus of grade Ⅱand above HCC, Which was positively correlated with the progress of the tumor grades, and the difference is very significant ③ Spearman correction analysis shew that P-Smad2 was negatively associated with that of PPM1A(r = -0. 345,P =0. 001). Conclusions the aberrant expression location and intensive of PPM1A and P-Smad2 and their interaction maybe take part in the pathogenesis of hepatocellular carcinoma together.
